Speculation points to something non-MGS.
Either way, I'm excited! I mean this guy's a genius. Yeah I know a lot of people think he's nuts or something but give the guy a break. Find someone who can handle the MGS better than him...
Itagaki? It will be full of boobs!
Miyamoto? The game might migrate to someone doing household chores.
Anybody from Capcom? Franchise will go without numbers anymore and have no existing exposition.
And so on...
I personally hope he does a horror game next. That way the cheesy dialogue and over the top angles would finally not be out of place.

1) Only teh PS3 haz the power / disk space / sixaxis for MGS4! 2) It's been out so long, no-one will buy it on 360 - just like no-one bought MGS2 on the XBOX! 3) You don't understand the honour of the Japanese culture (I'm an expert), it would be dishonourable for this to happen! 4) LOLZ! CRY MOAR! 5) etc.

Of course he kills people all the time! Only a genuine secret agent would know that you can totally distract a highly trained enemy combatant in the middle of a war-zone by throwing soft-core porn mags on the floor :)
Surely this: "Well, to give you maybe a surprise, people might think I love action games and I love shoot 'em ups and I kill people all the time, but that's not it." suggests that his next title will be decidedly un-actiony and almost totally non-shooty. So everyone hoping for ZOE3 will most likely be disappointed.
Otacon seemed to be used as a symbol for a female characters demise, which got a bit tired when pretty much everyone he knows and likes (sexually or otherwise) dies.
Don't get me wrong, I love the series, but some of the smaller story arcs are getting very tired, for me atleast.
@Nirach: I liked Raiden for what he was in both games. Personality kinda jumped insanely from 2 going into 4, but plot holes were filled just enough so I didn't fall and die. Besides, he fucking cuts muh fuggas up with super special sword-in-mouth-technique.
Rose - thought she bitched and moaned way too much. I liked that she was involved in the story for 4, but didn't like her for much else than being a useful plot device. Characterisation just wasn't there,in my opinion.
I liked Otacon's relationships because they (although they did indeed get tired toward the end) attempted showed a more human sense of despair you don't really see in games all that often. Was it a shame that ALL of the tragedy had to fall on him? Yeah. It weakened his character, but taken as a case by case basis, every relationship (and outcome) he had in the game was pretty... dare I say... solid.
All the Rose chats I couldn't skip lead to head+wall interaction for me. I actually have an urge to play it again now.
Admittedly the first ones were, as you say, solid. I just felt that as it continued to happen without relent, it watered down the whole thing. That, for me, detracted from the experience more than anything.
There were comedy games on pc, Monkey Island, Maniac Mansion, Leisure Suit Larry, tons came from Lucasarts. Warioware is also a very funny game and from more recent times Psychonauts was also pretty funny.

He really can talk about absolutely nothing for extended periods of time.
I'm not sure I'd want to see him do romance, especially if what was going on between Raiden and Rose was his idea of romance..

I liked Otacon's tragic romances. I will not comment on Raiden/Rose, however. Not that it was bad, just never liked Rose so much.
